Hello my O level equivalence was 86% and my A levels equivalence is 80%. But in A levels my least grade was an E which was in Biology. Now if I do well in the entry test and fall on merit, will the E in biology, which is an important subject to medical colleges, ruin my admission chances into private medical colleges like IMDC, Bahria etc.?

It's highly unlikely, as the universities only evaluate you based on your equivalence and Test scores, grades of individual subjects do not matter..
Are you an overseas student?

No I will apply on local seat in Private Medical Colleges. But I'm thinking that what if in the interview they bring this point up that your least score is in Bio, so that is what's worrying me a bit.

I don't think Interview plays a big role in the admission process, form what I know, The only think they want to check in an interview is your interest and your financial status (To make out if you'd be able to pay the fees.)
